1 About the contracting institution

The institution´s premises are mainly located in the Arrhenius laboratories together with the other chemistry institutions and other institutions in "Life Science". More than 200 people, including about 80 doctoral students, are currently employed by the department. The research spans a wide area and is within many of the projects of the highest world class. The department also has a strong connection to the Science for Life Laboratory. The research that is conducted is unique in Sweden and also in the world through its interdisciplinary nature and

2 Goals and purpose

The instrument should be used for semi-preparative and preparative procedures using mainly clinical samples. For future use the instrument should also be used to isolate or purify targets from complex solutions. The purpose of the purchase is to ensure continuous access to an FPLC instrument for the Nielsen laboratory team members.

3 Requirement specification

Describe the product or implementation of the service:

The instrument should offer reliable and effective means to in a semi-preparative and preparative way purify proteins and other biomolecules from simple or complex solutions including small volumes of clinical samples (≤500 uL). For this purpose a wide range of flow capacity is needed, preferably from very low 0.1mL/min up to 25 mL/min. The system should enable different chromatopgraphy (size exclusion, affinity-based purification) techniques and be compatible with the most common columns on the market. For our purpose the Superose 6 Increase 10/300 column is the most important tool.   

Product:

-The chromatography system must be CE marked

- The equipment must be compatible with the Swedish electrical system. The standard voltage is 230 V or 400 V and the standard frequency is 50 Hz

- Electric and electronic equipment must comply with the RoHs-directive regarding mercury, lead, hexavalent chromium and the flame retardants polybrominated diphenyl ethers (PDBE) and polybrominated biphenyls (PBBs)

- The instrument must have a inlet valve with a switch between multiple buffers.

- The instrument must have high performance dual piston pumps for flow rates from 0.1 to 25 ml/min and pressure up to 20 MP.

- The instrument must have a pressure sensor.

- The instrument must have a gradient mixer

- The instrument must have injection valve for sample application from loops and the flexibility to enable configuration modifications to allow sample application from a sample pump if needed in the future.

- The instrument must have UV/vis monitor for a fixed wavelength but the flexibility to modify the configuration to enable monitoring at multiple wavelengths simultaneously (190-700 nm) in the future.

- The instrument must have a light source that does not heat the sample during absorbance measurements

- The instrument must have flow cell for the UV/vis monitor with 2, 5 or 10 mm light path

- The instrument must have conductivity monitor for gradient verification

- All equipment must be delivered complete including all hardware to run the instrumentation as well as accessories

- The following spare parts or other parts must be included for the instrument:

Sample loop 500 µl, 1 ml and 2 ml - Injection kit

- All equipment must be delivered complete including all hardware and software to run the instrumentation as well as accessories.

- The instrument must have outlet valve for switching between a fraction collector, an outlet position and waste

- The instrument must have column valve for automatic cleaning using column bypass and reverse flow

- The instrument must have round fraction collector based on time, volume and peak recognition with drop synchronized function to decrease the spillage

- The instrument must have a fraction collector with the possibility to collect up to 175 fractions

- The instrument must have air sensor to pause the system if running out of buffer

- The instrument must have software, which allows automatic run of the system with modified gradients and including injection of samples and collection of fractures

- The instrument must be linked to a user friendly browser-based monitoring mechanism

- It must be possible to pause and resume elutions during runs

- The instrument must be able to operate in the cold (4-10 degrees centrigrade).

- Spare parts must be guaranteed for up to 5 years

- Delivery time must be a maximum 3 weeks

- Purchase must include installation and on-site user training for up to five lab members

- Complete documentation in English in handling and daily maintenance of the equipment must accompany the equipment.

- A user manual/instruction book installed on the computer supplied with the system (or accessible via the internet) which can be copied freely (or freely available from multiple computers).

- The documentation must contain: Instructions on how to minimize the environmental impact during installation, use, service and recycling/disposal, including instructions on how to minimize consumption of energy, water, consumable materials/parts and emissions in those cases when it is applicable.

- The documentation must contain: Recommendations on the proper maintenance of the product, including information on which spare parts can be replaced, cleaning advice

- The supplier will take full responsibility for installation, final adjustment, acceptance testing and commissioning of the tendered equipment, and assist in adapting the equipment to the laboratory environment in existing premises

- The warranty period of the equipment must be at least one year after successfully performed final acceptance test.

- Upgrade of software must be free during the warranty period

- Update of software should be free during the entire lifespan of the equipment

5. Evaluation and award (supplier selection)

The supplier offering the most advantageous solution to the contract is awarded the contract. SU evaluates the tenders in accordance with the above requirements.
