About Stockholm University

Stockholm University is characterized by openness and an innovative and cross-border activity in Sweden´s capital. Ranked as one of the world´s top 100 HEIs, Stockholm University is one of Europe´s leading institutions for higher education and research in human sciences and natural sciences.

Stockholm University was founded in 1878 with the ambition to renew the traditional universities in Sweden. Close contacts and an active exchange of experience and knowledge with the outside world characterize the business since its inception.

33,000 students, 1,600 doctoral students and 5,500 employees are active at Stockholm University. Within the science areas of science and human sciences, 190 study programs and 1,700 courses are offered. The university has a total turnover of SEK 4.93 billion.

Stockholm University contributes to knowledge development in a global perspective and in collaboration with others. This is done through education and research in the international front line, as well as in cross-border and multidisciplinary studies. In collaboration with the surrounding society, knowledge is made available to everyone through dialogue and participation in debate and social development.

1 About the contracting institution

The institution´s premises are mainly located in the Arrhenius laboratories together with the other chemistry institutions and other institutions in "Life Science". More than 200 people, including about 80 doctoral students, are currently employed by the department. The research spans a wide area and is within many of the projects of the highest world class. The department also has a strong connection to the Science for Life Laboratory. The research that is conducted is unique in Sweden and also in the world through its interdisciplinary nature and spans a very wide area including cell biology, biochemistry, biophysics and theory. For more information about our business, see our website www.dbb.su.se.

2 Goals and purpose

To produce 3D scientific animation for visualisation of research data aimed toward general academic community. The specific need is to visualise a molecular mechanism in a particular way, to provide suitable molecular context combining complex information for effective science communication.

3 Requirement specification

The project on the molecular visualization of the mechanism of membrane-tethered protein synthesis in mitochondria includes two-minute molecular animation of the principal aspects of the mechanism, including protein folding pathways comprising the gating mechanism.

Required experience: tomography and single-particle cryo-EM data handling in the MRC format, as well as atomic models in PDB. The related structural software that will be needed for preliminary analysis are Pymol, Chimera, Chimera-X. References for previous similar work

 will be required.

The following schedule is required.

- Concept & pre-production development – 1 week

Storyboard-Outline: initial written description of the sequence of events (scenes) in the visualization. Covers scientific concepts, planned visuals, and estimated timing of each scene.

Asset Preparation: pre-production analysis and processing of structure files for production phase.

Storyboard & Script: illustrated version of the Storyboard-Outline. Incorporates proxies or sketches of molecular actors and central concepts, initial color palette, composition and refined duration for each scene in the animation. Written Script to accompany English Voice-Over.

- Production – 5 weeks

• Draft Animation: animation created for feedback/evaluation. Aims to be close to final – it typically will not undergo changes that dramatically deviate from Storyboard.

- Final deliverable – 1 weeks

• Finished Animation (Final Art): MP4/H.264 codec at 1920x1080px, edited to accommodate feedback from phase 03. Delivered ready for use in manuscript, conferences, exhibits, web, and other settings.

- Outline (abbridged) duration: 2 min

1. Overview of the mitoribosome

2. Introduction to molecular actors principally involved and in relation to polypeptide tunnel

3. Conformational changes: mL45, uL23m, protein folding pathways comprising tunnel gating mechanism

4. Nascent chain: emergence in context of principal conformational changes; allowed path toward membrane 5. Nascent chain & Oxa1 insertase (interaction) in the context of mL45, uL23 conformational changes

6. Oxa1 / mitoribosome contact sites

7. Conclusion

The budget is restricted to 212 000 SEK.

4 Quotation

We expect quotation regarding the above and wish the price to be stated in the quotation.

Please state the total price for the assignment implementation. If budget exists, it can be specified for transparency.

No other costs may be added for the commission´s implementation.

Answer in the tender how the offered product meets the requirement specification:

The quotation will be submitted no later than 2021-01-05

5. Evaluation and award (supplier selection)

The supplier offering the most advantageous solution to the contract is awarded the contract. SU evaluates the tenders in accordance with the above requirements.
